11-Step Total Synthesis of (−)-Maoecrystal V

天然产物Maoecrystal V 具有独特的五环骨架,含有6个手性中心,其中包括4个连续手性中心,2个为连续手性的季碳。MV分子中具有并环、螺环、桥环、内酯环、四氢呋喃环等结构,对HeLa细胞系具有较强的选择性和细胞毒性(IC50 = 60 nM)。迄今为止,共有五个课题组完成了其全合成。Baran课题组基于生源合成假说,不使用D-A反应,而是采用Pinacol重排为关键步骤,以11步反应、2.7%的总收率完成了MV的不对称全合成,这对MV及其类似物的多样性合成和生物活性研究都具有重要意义。
